Overview & mechanism
SS-31 is a research designation associated with elamipretide. The selected studies concern primary mitochondrial myopathy, a defined disease population. They illustrate why promising early findings must be considered alongside later controlled trials rather than presented as a general enhancement claim. [1] [2]
Selected research
These study summaries preserve the type of evidence and the question being tested. They are a curated starting point, not a systematic review of every publication.
2018 · Early randomized trial
The early trial investigated safety and efficacy and reported exploratory exercise-capacity findings. [1]
Interpretation limit: A small early study is not a definitive estimate of efficacy or uncommon risks.
2023 · MMPOWER-3
The study did not demonstrate significant benefit on its primary walking-distance and fatigue endpoints. [2]
Interpretation limit: Secondary or subgroup findings should not be presented as if the primary trial had established efficacy.
How should an early signal and a negative primary result be reconciled?
Start with the prespecified endpoints of the larger trial. Exploratory analyses can generate questions, but they do not replace the main result. Differences in follow-up, participant characteristics and analysis methods may help explain divergent findings; they should be investigated rather than used to select only favourable numbers. A result in one disease setting also cannot establish benefit for healthy people.
A practical reading checklist
- Identify the model: cell culture, animal experiment or human study.
- Name the comparator: placebo, another intervention or an untreated control.
- Record the endpoint: what changed, how it was measured and at which time point.
- Read uncertainty: consider sample size, missing observations and confidence intervals, not only a headline percentage.
- Keep the conclusion within scope: a mechanism, an analytical result and a clinical outcome are not interchangeable.
Material identity before comparison
Ask whether the specification identifies the exact sequence, stereochemistry and chemical form. Research-code aliases help locate literature but do not establish equivalence between a catalogue vial and the clinical study product. If an assay depends on mitochondrial function, define the functional readout separately from chemical identity and purity: a mass-spectrum match is not a measurement of cellular performance.
Batch quality & COA
Read a report as a set of measurements on an identified sample. First match the product, lot, sample identifier and test date; then ask what each measurement actually establishes.
| Record | Question it addresses | What to avoid assuming |
|---|---|---|
| Identity and chemical form | Is the tested material consistent with the intended molecule? | A shared catalogue name establishes identical structure. |
| Chromatographic purity | What proportion of the measured signal is assigned to the main component under this method? | The percentage proves mass per vial, sterility or biological potency. |
| Content / assay | How much analyte was measured, using the stated method and reference? | The printed vial amount is itself an assay result. |
| Additional tests | Were relevant attributes, such as endotoxin, measured separately? | An unlisted test was performed or passed. |
| Lot and sample traceability | Does this document match the material under review? | A report for an earlier batch covers the next shipment. |
Retain the original report and its verification route. Record missing information as unknown rather than filling it in from a similar product. Read the COA verification guide.
Handling & research records
Use the handling instructions and safety documentation supplied for the exact material, presentation and lot. Do not copy a storage temperature or shelf life from a different formulation. Document receipt date, labelled storage requirements, container condition and any deviation for review under your laboratory's procedures.
Keep preparation records with the experimental dataset so that later results can be traced to a specific material. Appearance alone is not a stability measurement. If storage or shipping conditions are uncertain, request supporting information before treating the sample as suitable for the intended experiment. Explore storage documentation.
Common questions
Should MMPOWER-3 be omitted because earlier findings were encouraging?
No. A balanced summary includes its primary results and explains their study context.
Can a subgroup result override the main trial?
Not automatically. Prespecification, multiplicity and independent confirmation matter when interpreting subgroup analyses.
Does this guide describe all elamipretide indications or current approvals?
No. It reviews selected primary mitochondrial myopathy trials. Current authorization is indication- and jurisdiction-specific and is a separate question.
Does a research report establish safety for personal use?
No. Analytical quality, an experimental observation and clinical safety are different questions. This guide does not prescribe treatment or establish that research supplies are appropriate for human or veterinary use.

Check current trial registry records. Registry listings describe research records; a listing alone does not establish efficacy, approval or suitability of a supplied batch.
